PT-141 10 mg
Centrally-acting melanocortin analog · FDA-approved only for premenopausal women, never for men
Bremelanotide (PT-141) is a synthetic analog of the alpha-MSH hormone: it activates melanocortin receptors, mainly MC4R and MC1R, in the central nervous system. Unlike sildenafil and other PDE5 inhibitors, which act peripherally on penile blood flow, bremelanotide acts in the brain; the FDA's own label states that the exact mechanism by which it improves sexual desire in women 'is unknown' (FDA label for VYLEESI, section 12.1). The FDA approved it in June 2019 under the brand name Vyleesi, EXCLUSIVELY for acquired, generalized hypoactive sexual desire disorder (HSDD) in PREMENOPAUSAL women not due to another medical or psychiatric condition, relationship problems, or medication. The approved dose is 1.75 mg subcutaneously via autoinjector, given at least 45 minutes before sexual activity, with no more than one dose every 24 hours and no more than 8 doses per month. It was never approved for men or to enhance sexual performance (FDA label, sections 1 and 2.1).
